HincII is a type II restriction endonuclease which binds and cleaves duplex DNA at sequences GTY|RAC (Y=C or T, R=A or G, | denotes site of cleavage, numbering is G1T2Y3R4A5C6). The three dimensional x-ray crystal structure shows that HincII (green, salmon, below) distorts its bound DNA (blue, below), intercalating the side chain of residue Gln 138 (pink, below) into the duplex just outside of the six base pair recognition sequence (Horton, et al. (2002) Nat. Struct. Biol. 9, 42-7).
